Radiopharmaceutical kit to prepare Copper-64 labelled porphyrin-lipid nanoparticles for radiotracer imaging studies in cancer patients

Abstract
Porphyrin-lipid nanoparticles (porphysomes) are intrinsically multimodal nanoparticles with promising applications in cancer theranostics. Here, we report the development of a radiopharmaceutical kit for the facile preparation of copper-64 (64Cu) labelled porphyrin-lipid nanoparticles for PET imaging in cancer patients. The kit enables rapid, one-step radiolabeling of porphyrin-lipid nanoparticles with 64Cu under mild conditions with high radiochemical yield and purity. The radiolabeled nanoparticles demonstrated excellent stability in serum, favorable biodistribution profiles, and selective tumor accumulation in xenograft models. First-in-human PET imaging studies confirmed tumor-selective uptake and delineation, establishing the radiopharmaceutical kit as a practical platform for clinical translation of porphyrin nanoparticle-based PET imaging in cancer patients.
